Fig.1 Micro-CT analysis of skeleton in 21-day-old WT C57BL/6 mice and HE Fgfr3-G374R mice.
Abbr. WT, wild type; HE, heterozygous.

Fig.2 Micro-CT analysis of skull in 21-day-old Fgfr3-G374R mice.
Significant cranial rounding can be observed in all heterozygous Fgfr3-G374R mice, as indicated by the red circle.
Abbr. WT, wild type; HE, heterozygous.

Fig.3 Analysis of body and various bones length in 21-day-old Fgfr3-G374R mice.
The results revealed significant reductions in R-Op distance in both sexes and in foramen magnum dimensions (length and width) in male Fgfr3-G374R mice, compared to WT controls (n=5-12, Kolmogorov–Smirnov test or Unpaired t-test, *P<0.05, **P<0.01, ***P<0.001, ****P<0.0001, ns: no significance).
Abbr. WT, wild type; HE, heterozygous; R-Op distance, the linear distance from rhinion to opisthion.

Abbr. WT, wild type; HE, heterozygous.

Fig.5 Micro-CT analysis of skull in 36-day-old Fgfr3-G374R mice.
Significant cranial rounding can be observed in all heterozygous Fgfr3-G374R mice, as indicated by the red circle.
Abbr. WT, wild type; HE, heterozygous.

Fig.6 Analysis of body and various bones length in 36-day-old Fgfr3-G374R mice.
The results revealed a shortening of the body, tibia, R-Op distance and foramen magnum length in both sexes of mutant mice. Femoral length was reduced exclusively in mutant males and foramen magnum width was diminished in mutant females (n=5-12, Kolmogorov–Smirnov test or Unpaired t-test, *p<0.05, **p<0.01, ***p<0.001, ****p<0.0001, ns: no significance).
Abbr. WT, wild type; HE, heterozygous; R-Op distance, the linear distance from rhinion to opisthion.

Fig.7 Representative photographs showing that Fgfr3-G374R mice exhibited a slower growth phenotype compared with WT littermates (n=3, 63 days old).
Abbr. WT, wild type; HE, heterozygous.

Fig.8 Analysis of body weight, brain weight and body lenght of 63-day-old Fgfr3-G374R mice.
The results revealed significant reductions in body weight and body length in male Fgfr3-G374R mice, as well as nose-tail length in female Fgfr3-G374R mice, compared with WT controls. In contrast, brain weight was consistently greater in both male and female Fgfr3-G374R mice, likely reflecting the constitutive activation of FGFR3 caused by the mutation (n=5-12, Kolmogorov–Smirnov test or Unpaired t-test, *P<0.05, **P<0.01, ***P<0.001, ****P<0.0001, ns: no significance).
Abbr. WT, wild type; HE, heterozygous; R-Op distance, the linear distance from rhinion to opisthion.
