Fig.1 Detection of EDNRA expression in lung and heart by RT-PCR. Lung and heart RNA were extracted from male 4-week-old wild-type SD rats (WT, n=2) and homozygous hEDNRA(SD) knockin rat (HO, n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Rat Ednra mRNA (191 bp) was detectable only in wild-type rat. Human EDNRA mRNA (214 bp) was detectable only in homozygous hEDNRA(SD) knockin rat but not in wild-type rat.

Fig.2 Detection of EDNRA expression in hEDNRA(SD) rats by WB. Tissue lysates of the heart and lung were collected from wild type SD rats (WT, male, 6 week old) and homozygous hEDNRA(SD) rats (HO, male, 6 week old), and then analyzed by western blot with anti-EDNRA antibody. 40 μg of total proteins were loaded for western blotting analysis. The antibody is cross-reactive between human and rat EDNRA.
