Fig.1 Detection of MAPT expression in cerebellum by RT-PCR.
Wild type: only one band at 200 bp with primers F1/R1 (mMapt);
Homozygous: only one band at 181 bp with primers F2/R2 (hMAPT);
Abbr. M, DNA marker; HO, homozygous; WT, wild type.

Fig.2 Detection of MAPT expression in cerebellum by RT-PCR. Cerebellum RNA was extracted from 11-12-week-old male wild-type C57BL/6 (WT)(n=3) and 11-14-week-old male homozygous hMAPT knockin mice (HO) (11w: n=2, 12w: n=3, 13w: n=2, 14w: n=1), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Human MAPT mRNA (2079 bp) was detectable only in homozygous hMAPT knockin mice but not in wild-type mice.
Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.3 Detection of MAPT expression in cerebellum by qPCR. Cerebellum RNA was extracted from 11-12-week-old male wild-type C57BL/6 (WT)(n=3) and 11-14-week-old male homozygous hMAPT knockin mice (HO) (11w: n=2, 12w: n=3, 13w: n=2, 14w: n=1), then cDNA libraries were synthesized by reverse transcription, followed by qPCR with human MAPT and mouse Gapdh primers. Relative expression represents the human MAPT mRNA level relative to its average expression in homozygous hMAPT knockin mice. Human MAPT mRNA was detectable only in homozygous hMAPT knockin mice but not in wild-type mice.
Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.4 Detection of MAPT expression in cerebellum by qPCR. Cerebellum RNA was extracted from 11-12-week-old male wild-type C57BL/6 (WT)(n=3) and 11-14-week-old male homozygous hMAPT knockin mice (HO) (11w: n=2, 12w: n=3, 13w: n=2, 14w: n=1), then cDNA libraries were synthesized by reverse transcription, followed by qPCR with mouse Mapt and mouse Gapdh primers. Relative expression represents the mouse Mapt mRNA level relative to its average expression in homozygous hMAPT knockin mice. Mouse Mapt mRNA was detectable only in wild-type mice but not in homozygous hMAPT knockin mice.
Abbr. M, marker; HO, homozygous; WT, wild type.

Fig 5. Detection of mouse MAPT expression in Brain in hMAPT knockin mice by IHC.
Brains were collected from wild-type mice (+/+) and homozygous hMAPT mice (HO), and analyzed by IHC with anti-human MAPT antibody.
Abbr. HO/HO, homozygous; +/+, wild type.
Note. The anti-mouse MAPT Antibody cross-reacted with mouse MAPT and humanized MAPT.

Brains were collected from wild-type mice (+/+) and homozygous hMAPT mice (HO), and analyzed by IHC with anti-human MAPT antibody.
Abbr. HO/HO, homozygous; +/+, wild type.
Note. The anti-human MAPT Antibody cross-reacted with mouse MAPT and humanized MAPT.

Fig 7. Detection of human MAPT expression in Brain in hMAPT knockin mice by IF.
Brains were collected from wild-type mice (+/+) and homozygous hMAPT mice (HO), and analyzed by IF with anti-human MAPT antibody.
Abbr. HO/HO, homozygous; +/+, wild type.
Note. The anti-human MAPT Antibody cross-reacted with mouse MAPT and humanized MAPT.
