Model Description

Exon 1-3 of Csf2 gene was deleted to generate Csf2 knockout mice. Homozygotes for targeted null mutations exhibit lung abnormalities with lymphocytic infiltration and accumulation of surfactant lipids. Litter sizes from homozygous breeding pairs are smaller at weaning due to perinatal mortality (Note from MGI).

Fig.1 Detection of Csf2 expression in lung by RT-PCR. Lung RNA was extracted from 7-year-old male wild-type C57BL/6 (WT) and homozygous Csf2-KO mice (HO) (n=3), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Mouse Gapdh mRNA (123 bp) was detectable both in wild-type C57BL/6 mice and homozygous Csf2-KO mice. Mouse Csf2 mRNA (146 bp/138 bp) was detectable only in wild-type mice but not in homozygous Csf2-KO mice.

Fig.3 Detection of mouse CSF2 expression in serum of WT C57BL/6 mice and HO CSF2-KO mice by ELISA. Serum were collected from wild-type C57BL/6 mice (n=3, male, 11 weeks old) and HO CSF2-KO mice (n=3, male, 11 weeks old), and analyzed by ELISA with anti-mCSF2 ELISA kit. Mouse CSF2 was exclusively detectable in wild-type C57BL/6 mice.
