谱系示踪


常用的谱系示踪是将特定启动子驱动的 Cre 工具小鼠和 Rosa26-LSL-Reporter 报告基因小鼠结合使用。特异性表达的 Cre 酶会切除 loxP 位点之间的转录终止序列,从而激活报告基因的持续表达。由于该修饰在 DNA 水平上进行,可以遗传到子代细胞,因而可以永久标记特定类型的细胞。通过观察这些被标记上的荧光信号来追踪某类特定细胞及其后代所有细胞的增殖、分化以及迁移等活动,为研究细胞命运及谱系提供有力手段。如下图所示:
