KPC

Strain Name:C57BL/6Smoc-Trp53em4(R172H)Krasem4(LSL-G12D)Tg(Pdx1-cre)Smoc
Strain State:Repository Live | Cat. NO.:NM-KI-210096

The KPC mouse is an established and clinically relevant model of pancreatic ductal adenocarcinoma (PDAC) which develops many key features observed in human PDAC including pancreatic intraepithelial neoplasia alongside a robust inflammatory reaction including exclusion of effector T cells. Metastases are observed in around 80% of KPC animals located primarily in the liver and lungs. Mutations in both KRAS and TP53 genes are found in around 80% and 70% of all human PDACs respectively. Trp53-R172H、Kras-LSL-G12D were crossed with Pdx1-Cre-Tg to generate KPC mice. The KPC mouse contains a dominant negative point mutation in the transformation related protein 53 gene (TP53R172H), and a conditional activation point mutation in the KRAS gene (KRASG12D). A lox-stop-lox termination sequence is encoded upstream of KRAS mutated genes to prevent expression in the absence of Cre recombinase. The pancreas-specific Pdx-1 promoter enables expression of Cre recombinase in acini, islet and duct cells of the pancreas. Cre-mediated recombination excises the lox-stop-lox termination sequences and enables expression of KRASG12D in pancreatic tissue.

Mice

Pdx1-2A-CreERT2

Strain Name:C57BL/6Smoc-Pdx1em1(Avi tag-2A-CreERT2)Smoc
Strain State:Repository Live | Cat. NO.:NM-KI-18042

An Avi tag-2A-CreERT2 cassette was inserted at the Pdx1 gene stop codon site .

Mice

Pdx1-2A-Cre

Strain Name:C57BL/6Smoc-Pdx1em(2A-Cre)1Smoc
Strain State:Sperm cryopreservation | Cat. NO.:NM-KI-225082

2A-Cre expression cassette was knocked into the Pdx1 gene .

Mice

Pdx1-Flox

Strain Name:C57BL/6Smoc-Pdx1em1(flox)Smoc
Strain State:Developing | Cat. NO.:TBD

These mice carry loxP sites flanking target exons of Pdx1 gene. When crossed with a Cre recombinase-expressing strain, this strain is useful in eliminating tissue-specific conditional expression of Pdx1 gene.

Mice

Pdx1-KO

Strain Name:C57BL/6Smoc-Pdx1em1Smoc
Strain State:Sperm cryopreservation | Cat. NO.:NM-KO-226397

exon 2 of Pdx1 gene was deleted to generate Pdx1 knockout mice.

Mice

KPC(2)

Strain Name:C57BL/6Smoc-Trp53tm7(R270H-flox)Krasem4(LSL-G12D)Tg(Pdx1-cre)Smoc
Strain State:Repository Live | Cat. NO.:NM-KI-220014

Trp53-LSL-R270H, Kras-LSL-G12D were crossed with Pdx1-Cre-Tg to generate KPC mice. While KPC mice function similarly to KPC(2) mice, for more detailed information please contact our technical advisor.

Shanghai Model Organisms Center provides multiple types of tumor-bearing mouse models for cancer research and drug efficacy testing, including cell line-derived xenograft model (CDX) and patient-derived xenograft model (PDX). While PDX models better reflect the heterogeneity and diversity of human cancers, CDX models allow researchers to advance their pre-clinical drug development in a cost- and time-efficient manner.

research on cancer cancer model mouse models of cancer

The laboratory mouse is widely considered as a valid and affordable model organism for studying human disease. Mouse models of cancer not only enable researchers to understand the genetic basis of tumor development in complicated and dynamic physiological systems, but also serve as irreplaceable tools for developing and testing new therapies.  Shanghai Model Organisms Center offers a variety of mouse models of cancer to meet the needs of both academic and industry partners.
