Model Description

This Alpl knockout mouse model was obtained by knocking out the exon 3-4 region of the Alpl gene.

Validation Data

Fig.1 Survival of Alpl-KO mice in postnatal 15 days. (n=24 for the WT C57BL/6 group, n=50 for the HE Alpl-KO group, n=24 for the HO Alpl-KO group). (Data provided by a collaborator)

Continuous supplementation of vitamin B6 in the diet from the pregnancy stage can significantly reduce the mortality rate caused by epilepsy in homozygous individuals.

Abbr. WT, wild type; HE, Heterozygous; HO, Homozygous.

Fig.2 Body weight of Alpl-KO mice.

The body weight of homozygous Alpl-KO mice was markedly lower than that of both WT and heterozygous Alpl-KO mice (n=24 for the WT C57BL/6 group, n=50 for the HE Alpl-KO group, n=24 for the HO Alpl-KO group. Mean ± SEM. Two-way ANOVA , **p < 0.01, ***p < 0.001, ****p < 0.0001). (Data provided by a collaborator)

Abbr. WT, wild type; HE, Heterozygous; HO, Homozygous.

Fig.3 Bone length of Alpl-KO mice.

The result showed a significant reduction of the bone length in homozygous Alpl-KO mice compared with WT C57BL/6 mice at the age of 1.7-2.1 weeks (n=3 in WT C57BL/6 group, n=7 in HO Alpl-KO group. Mean ± SEM. t-test, **p < 0.01, ***p < 0.001, ****p < 0.0001). (Data provided by a collaborator)

Abbr. WT, wild type; HO, Homozygous.

Fig.4 Body length of Alpl-KO mice.

The body length (A) and the body length gain (B) from postnatal days 7 to 14 were both significantly decreased in homozygous Alpl-KO mice compared to WT C57BL/6 mice (n=24 for the WT C57BL/6 group, n=50 for the HE Alpl-KO group, n=20 for the HO Alpl-KO group. Mean ± SEM. Two-way ANOVA for A and One-way ANOVA for B, *p < 0.05, ****p < 0.0001, ns: no significance). (Data provided by a collaborator)

Abbr. WT, wild type; HE, Heterozygous; HO, Homozygous.
