Gene Summary

Gene Symbol
Stab1

Model Description

The endogenous mouse Stab1 gene was replaced by human STAB1 gene.

Validation Data

Fig.1 Detection of STAB1 expression in lung by RT-PCR.

Wild type: only one band at 147 bp with primers F1/R1(mStab1);

Heterozygous: one band at 147 bp with primers F1/R1(mStab1) and one band at 206 bp with primers F2/R2(hSTAB1).

Abbr. M, DNA marker;  HE, heterozygous; WT, wild type.

Fig.2 Detection of STAB1 expression in ovary by RT-PCR. Mouse Stab1 mRNA (147 bp) was detectable only in wild-type C57BL/6 mice. Human STAB1 mRNA (206 bp) was detectable only in homozygous hSTAB1 knockin mice but not in wild-type mice. Tissues RNA was extracted from a 8-week-old female WT C57BL/6 mouse and a 6-week-old female homozygous hSTAB1 knockin mouse, then cDNA libraries were synthesized by reverse transcription, followed by PCR.

Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.3 Detection of human STAB1 expression in hSTAB1 knockin mice by IHC. Human STAB1 was exclusively detectable in mesenteric lymph nodes and spleen tissues from homozygous hSTAB1 mice but not in WT mice. Tissues were collected from 12-week-old male WT C57BL/6 mice (n=2) and 11-week-old male HO hSTAB1 knockin mice (n=2), then analyzed by IHC. Black arrows indicate positive staining (brown). The scales are same in each image. Magnification, 20X.

Abbr. HO, homozygous; WT, wild type.
