Tissue RNA was extracted from female 7-week-old wild-type C57BL/6 mice (WT) and homozygous hSEZ6 knockin mice (HO), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human mRNA primers. Mouse Sez6 mRNA (451 bp) was detectable only in wild-type C57BL/6 mice. Human SEZ6 mRNA (441 bp) was detectable only in homozygous hSEZ6 knockin mice but not in wild-type mice.

Tissue lysates of hippocampus and cortex were collected from 7-week-old female wild-type C57BL/6J mice (WT) and homozygous hSEZ6 mice (HO), and then analyzed by western blotting with anti-SEZ6 antibody (Abcam, ab314233). 20 μg of total proteins were loaded for western blotting analysis.
The antibody was cross-reactive between human and mouse SEZ6.
