Fig.1 Detection of DLK1 expression in in mammary gland and adrenal gland by RT-PCR. Tissues RNA was extracted from 7-8-week-old female WT C57BL/6 mice and homozygous hDLK1 knockin mice (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Human DLK1 mRNA (175 bp) was exclusively detectable in homozygous hDLK1 knockin mice but not in WT mice, whereas muman Dlk1 mRNA (146 bp) was exclusively detectable in WT mice but not in hDLK1 knockin mice.

Fig.2 Detection of DLK1 expression in hDLK1 knockin mice by WB. Lysates of liver, adrenal gland and mammary gland tissues were collected from 7-8-week-old female WT C57BL/6 mice and homozygous hDLK1 knockin mice, then analyzed by western blot with anti-DLK1 antibody. DLK1 protein was obviously detectable in adrenal gland tissues from both adult WT C57BL/6 mice and homozygous hDLK1 knockin mice, with a trend toward an increase in the humanized mice, although the antibody was cross-reactived between human and mouse DLK1.
