Fig.1 Detection of DKK1 expression in thymus by RT-PCR. Thymus RNA was extracted from 11-week-old male WT C57BL/6 and homozygous hDKK1 knockin mice (n=3 in each group), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Mouse Dkk1 mRNA (426 bp) was detectable only in wild-type C57BL/6 mice. Human DKK1 mRNA (220 bp) was detectable only in homozygous hDKK1 knockin mice but not in wild-type mice.

Fig.2 Detection of human DKK1(A) and mouse DKK1(B) expression in serum of WT C57BL/6 mice and HO hDKK1 mice by ELISA. Serum samples were collected from wild-type C57BL/6 mice (n=3, male, 11 weeks old) and HO hDKK1 mice (n=3, male, 11 weeks old), and analyzed by ELISA with anti-mDKK1 ELISA kit and anti-hDKK1 ELISA kit. Human DKK1 protein was exclusively detectable in homozygous hDKK1 knockin mice, while mouse DKK1 protein was exclusively detectable in wild-type C57BL/6 mice.
